The church, built to plans by architect Francesco Fornari, completed and consecrated in 1968, is the seat of the parish of the same name, established on 19 May 1954 by decree of Cardinal Vicar Clemente Micara Neminem fugit. It is dedicated to the first martyrs of the Roman Church, who died during the persecutions unleashed by Emperor Nero in Rome, starting in July 64 and continuing until 67.
Since 1969 it has been the seat of the cardinal title of the Holy Protomartyrs in Via Aurelia Antica.
The church is preceded by a portico divided by pillars, under which is the dedicatory inion: D.O.M. in honorem SS. Protomartyrum. A.D. MCMLXVIII.
